TriAltus optimized Plasmid #10 for the purification of membrane proteins. This plasmid includes an N-terminal His8 tag and a C-terminal CL7 tag. Calnexin is encoded within the plasmid as a model membrane protein, inserted at BamHI/SpeI restriction sites.
Transcription is driven by the T7 RNA polymerase, and the plasmid is designed for expression in E. Coli.
This plasmid contains a C-terminal CL7 tag that is downstream of both a PreScission (PSC) and Sortase A (SRT) cleavage site.
Plasmid #10 contains two SRT cleavage sites, one downstream of the His8 tag before the model protein and one upstream of the CL7 tag and after the PSC cleavage site. The PSC cleavage site follows an 82-amino acid linker region downstream of the model protein.
