Plasmid #8 is used for the expression of membrane proteins using an engineered signal peptide. It includes a C-terminal His8 tag and CL7 tags, and also encodes an engineered signal peptide from the Bacillus halodurans YidC gene. There are two cleavage sites: one before the His8 tag and another in between the His8 tag and the CL7 tag.

EXPRESSION
Transcription is induced with IPTG and driven by the T7 RNA polymerase. The plasmid is designed for expression in E. coli

PEPTIDE TAG
Plasmid #8 contains a C-terminal His8 tag and CL7 tag that is downstream from a PreScission Protease cleavage site. A SUMO protease cleavage site sits in between the C-terminal His8 and CL7 tag.

CLEAVAGE SITE(S)
A SUMO protease cleavage site is located in between the C-terminal His8 tag and the CL7 tag, which would allow your protein to retain the His8 tag while using SUMO protease to cleave your protein off the column to get rid of the CL7 tag. Additionally, a PreScission protease cleavage site is located upstream of the His8 tag giving you the option to have tag-free protein.
