The challenge
A slow, low-yield process for a critical enzyme.
The client produced ultra-pure phi29 in-house with a purification that took four days and four chromatography steps, and yielded 0.25 mg of protein per gram of cells. For an enzyme at the heart of a diagnostic product, that process set the cost and pace of every lot.
What we did
One CLīM step instead of four.
We manufactured the same ultra-pure phi29 DNA polymerase using CLīM affinity purification, replacing the multi-column process with a single chromatography step completed in one day.
One affinity step replaces four chromatography steps
Purification in one day instead of four
Ultra-pure enzyme for a diagnostic application
Less buffer, water and energy per lot
Results
Better manufacturing economics.
12× higher yield
3 mg of protein per gram of cells, versus 0.25 mg with the client’s internal process.
75% less downstream processing
One chromatography step in one day, versus four steps over four days.
>50% less water and energy
Fewer columns and less time mean less buffer, water and energy for every lot.
Why it matters
What this means for your product.
1
Lower cost of goods
Higher yield and fewer steps reduce the cost of every lot of enzyme.
2
Faster turnaround
A one-day purification shortens lead times and makes it easier to respond when demand changes.
3
A leaner supply chain
Less water and energy per lot supports your own sustainability goals.
Comparison is against the client’s internal process for ultra-pure phi29 DNA polymerase. Yields are mg of purified protein per gram of cells.
