This plasmid contains the mtRNAP protein – a model for mitochondrial DNA/RNA-binding multisubunit proteins – with a C-terminal CL7 tag.
Transcription is driven by the T7 RNA polymerase, and the plasmid is designed for expression in E. Coli.
The CL7 tag is fused to the C-terminus of the third subunit of the mtRNAP protein.
Plasmid #2 contains several PreScission protease (PSC) sites: one at the N-terminus of subunit 2 (RpoB) and five within the third subunit-coding section (one at the N-terminus of RpoC, two at the N-terminus of RpoZ, and two at the C-terminus of RpoZ).
The plasmid includes a Trx tag at the N-terminus of subunit 2 and a NusA tag upstream of subunit 3.
